Design Notes

The trans element is an activating riboregulator which binds to a complimentary region on the cis repressor and opens up the RBS of the previously-repressed gene. When a plasmid containing this construct is present in a system that has the appropriate cis-repressed gene, activation of that gene ensues. Trans1 activates cis1, cis2, cis3, and cis4 Design specifics: free energy = -22.6kcal; 4bp loop, 2 bulges, 15bp tail



Source

The pBad and TT portions of the sequence came from the pCS958 template from the Smolke lab. The ta1 region was designed as an insert between these regions.
